PRINCIPLE OF THE METHOD

 

Urea/ Indole test는 요소 분해 효소와 인돌 생성을 기반으로 미생물을 동정하는 데 사용되는 키트입니다. 요소 분해 효소를 생성하는 미생물은 요소를 암모니아, 물, 이산화탄소로 가수분해 하며 알칼리성이 되면 Phenol Red 지시약에 의해 색상이 노란색에서 분홍색으로 변하게 됩니다. Tryptophanase를 생성하는 미생물은 트립토판을 인돌, 피루브산, 암모니아로 가수분해하며 Indole이 존재하게 되면 Kovac’s reagent와 반응하여 시험관 표면에 붉은색을 띄게 합니다.

 

 

 

PROCEDURE

 

• 키트를 꺼내어 실온에 잠시 방치합니다.

• 멸균된 루프를 사용하여 순수 배양된 의심 집락을 튜브에 접종합니다

• Vaseline oil을 2~3방울 첨가합니다.

• 35±2℃에서 1~4시간에서 최대 24시간 동안 배양 후 Urease 검사 결과를 확인합니다.

• Kovac’s reagent를 2~3방울 넣고 Indole 검사 결과를 확인합니다.

 

 

 

STORE

 

Store the kit at 2 - 6°C in its original package tightly closed and use it before the expiry date on the label. Keep away from sources of heat and avoid excessive changes of temperature.
